In plants and algae, light-dependent reactions occur in the thylakoid membranes inside chloroplasts. “Chloroplast” names the organelle; “thylakoid membrane” identifies the more precise location.
The membrane holds the photosystems, electron carriers, and ATP synthase needed to convert light energy into chemical energy. Water splitting supplies electrons and releases oxygen. Electron transport helps build a proton gradient across the membrane; ATP synthase uses proton flow back toward the stroma to produce ATP. NADP+ is reduced to NADPH on the stromal side.
Membrane, lumen, and stroma
- Thylakoid membrane: the main site of the light-driven electron transport machinery.
- Thylakoid lumen: the enclosed space where protons accumulate.
- Stroma: the surrounding fluid, where the Calvin cycle uses ATP and NADPH.
The answer is therefore not simply “the stroma.” Nor does it mean all photosynthetic organisms possess chloroplasts: cyanobacteria are prokaryotes and have a different cellular organization.
